摘要: Human stem cells are powerful tools by which to investigate molecular mechanisms of cell growth and differentiation under normal pathological conditions. Hedgehog signaling, the dysregulation causes several pathologies, such as congenital defects cancer, is involved in processes interferes with adipocyte rodent cells. The present study was aimed at investigating effect pathway modulation on phenotype using different sources human mesenchymal cells, bone marrow stromal multipotent adipose-derived We bring evidence that signaling decreases during differentiation. Inhibition this not sufficient trigger adipogenesis, but activation alters morphology well insulin sensitivity. Analysis glycerol-3-phosphate dehydrogenase activity expression marker genes indicate purmorphamine impairs adipogenesis. In sharp contrast reports maturation process, early steps differentiation, affected activation. targeting CCAAT enhancer-binding protein α peroxisome proliferator-activated receptor (PPAR) γ2 expression, whereas PPARγ1 level remains unaffected. Although stimulation does modify total number adipocytes, adipogenesis appears dramatically impaired, reduced lipid accumulation, a decrease adipocyte-specific markers, acquisition an insulin-resistant phenotype. This indicates necessary unveils striking difference process between cells. Disclosure potential conflicts interest found end article.
