Ectopic Expression of Gamma Interferon in the Eyes of Transgenic Mice Induces Ocular Pathology and MHC Class II Gene Expression

摘要: Purpose. To direct the expression of 7IFN to eyes transgenic mice as a means investi-gating possible role this lyinphokine in ocular paihogenesis.Methods. Transgenic mouse strains were generated by injection o DNf A fragment containingthe murine aA-crystallin promoter fused coding sequence gene. PCRand RT-PCR used screen for presence transgene and 111RNA analyses,respectively. Methacrylate-embedded eye sections analyzed morphology cryosec-tions immunoperoxidase antibody staining.Results. The most notable effects these include cataract, mi-crophthalmia, blepharophimosis, microphakia, impairment lens fiber formation, arrest ofretinal differentiation, serous retinal detachment with macrophages subreti-nal space, persistent, hyperplastic primary vitreous, corneal vascularization. MHC class IIniRNA levels significantly increased II proteinswere expressed their cornea, iris, ciliary body, choroid, RPF...Conclusions. Fxiopic dec ted growth whole eye,resulting microphthalmia microphakia. authors' data suggest that cvACry-7lFNtransgenic cells express functional receptors interaction 7IFNwith its receptor induced biochemical morphologic changes eyes.
