摘要: The centromere is the chromosomal region in which kinetochore assembled to orchestrate chromosome segregation. It defined by presence of a histone H3 variant called Centromere Protein A (CENP-A) or CenH3. Propagation identity entails deposition new CENP-A upon exit from mitosis vertebrate cells. group 16 proteins that co-immunoprecipitate with CENP-A, Constitutive Associated Network CCAN, contribute assembly and function. For most them it still unclear how when they are recruited centromeres whether have role deposition. Taking advantage Xenopus egg cell-free system, we addressed these issues for CCAN CENP-C, CENP-T CENP-W. CENP-C recruitment occurs as soon sperm DNA, containing added extract, continues after de novo incorporation early interphase. In contrast, centromeric late interphase precedes CENP-W, mitosis. Unlike CENP-W do not participate However, like play major assembly. Depletion results reduced amount at centromeres, an effect more prominent than spite this, kinetochores can be under this condition although Ndc80 Mis12 decreased. Our support existence 2 pathways directed CENP-T/W, reconstituted extracts.
