Monoclonal antibodies detecting antigenic determinants with restricted expression on erythroid cells: from the erythroid committed progenitor level to the mature erythroblast.

摘要: Two new cell surface antigens specific for the erythroid lineage were defined with cytotoxic IgM monoclonal antibodies (McAb) (EP-1; EP-2) that produced using BFU-E-derived colonies as immunogens. These two are expressed on in vivo and vitro derived adult fetal erythroblasts, but not erythrocytes. They detectable resting lymphocytes, concanavalin-A (Con-A) activated lymphoblasts, granulocytes, monocytes or granulocytic cells macrophages present peripheral blood harvested from CFU-GM cultures. Cell line tissue distributions distinguish McAb EP-1 EP-2 all previously described antibodies. (for erythropoietic antigen-1) inhibits formation of BFU-E CFU-E, CFU-GM, complement-dependent cytotoxicity assays. By sorting analysis, about 90% progenitors (CFU-E, BFU-E) recovered antigen-positive fraction. Seven percent this fraction (versus 0.1% negative fraction). The expression antigen is greatly enhanced K562 cells, inducers hemoglobin synthesis. fails to inhibit CFU-E colony predominantly immature it weakly mature erythroblasts. findings provide evidence (BFU-E CFU-E) express determinants fail be other progenitor hence appear unique lineage. potentially useful studies differentiation isolation.
