摘要: In this study changes in the amount and distribution of types I III collagen mRNA protein were investigated rat heart after induction a left ventricular myocardial infarction (MI). Sham operated rats served as controls. The animals sacrificed at different time intervals operation. Northern blotting cardiac RNA hybridization with cDNA probes for procollagen revealed 5- to 15-fold increase infarcted ventricle. Type levels already increased day 2 MI, whereas type followed response 4 MI. This was sustained least 21 days ventricle mRNA, 1 still elevated 90 noninfarcted right 7-fold observed both levels, but only non-infarcted septum transient from 7-21 (4- 5-fold increase) decline sham thereafter. 7 MI compared situ same showed mRNA-producing cells area around necrotic cardiomyocytes, interstitial part myocardium. No labeling detected above cardiomyocytes. Combined immunohistochemistry that producing have myofibroblast-like phenotype myocardium are fibroblasts by an deposition, measured computerized morphometry on sirius red-stained tissue sections well hydroxyproline assay. collagen-positive maximal 14 (3- controls) slightly declined 21. 57 +/- 10% Hydroxyproline contents significantly septum.(ABSTRACT TRUNCATED AT 400 WORDS)
