摘要: Delayed graft function remains an important complication after renal transplantation. In this study, we investigated the influence of trimetazidine (TMZ), a cytoprotective agent, on medullary damage prolonged preservation and autotransplantation. Pig kidneys were cold-flushed preserved (48 h at 4°C) with two standard solutions Euro-Collins University Wisconsin supplemented or not TMZ (10−6 M). Analysis plasma urine from 48-h-cold-stored autotransplanted was performed biochemical methods proton NMR spectroscopy. Histological study by light electron microscopy reperfusion (30–40 min) day 14. The results showed that in either solution containing improved significantly glomerular filtration rate compared without TMZ. reduced damage, evidenced decreased excretion trimethylamine- N -oxide, dimethylamine, dimethylglycine, acetate urine. Proximal tubular injury TMZ-free groups assessed greater Na+ excretion, amino aciduria, lactic aciduria than TMZ-supplemented groups. Urinary concentrating ability TMZ-preserved groups, there also citrate, which is citric acid cycle metabolite. An extensive reduction apical brush border cells, notably those proximal tubules, noted This clearly shows has beneficial action vivo its major impact vulnerable medulla.
