Fixed-bed dialysis culture of a transfectoma cell line producing chimeric Fab-fragments with “nutrient-split”-feeding strategy

摘要: The very shear sensitive transfectoma cell line, FAB 10, producing a chimeric Fab-antibody fragment specific for the human carcinoembryonic antigen (CEA), was cultivated in fixed bed reactors where cells were immobilized macroporous carriers. As line had low productivity, cultures with “process integrated product enrichment” performed membrane dialysis bioreactor bed, molecular weight metabolites are removed over and high products enriched. In new “nutrient-split” feeding strategy concentrated medium supplied directly to unit, whereas buffer solution used as fluid. With technique up 5800 μg Fab l−1 could be obtained, more than 10 times higher compared without or batch suspended cells. “nutrient-split”-feeding reduced amount of required per mg antibody significantly. © Rapid Science Ltd. 1998
