摘要: Abstract Carcinoembryonic antigen (CEA) is one of the best characterized tumor-associated antigens and extensively used in vitro immunodiagnosis human colon adenocarcinomas. Among a number anti-CEA monoclonal antibodies, murine antibody T84.66 shows highest specificity affinity for CEA has been successfully vivo tumor imaging mice humans. We report here cloning sequencing genes coding amino acid sequence variable regions heavy light chains. also construction mouse/human chimeric IgG1 using region constant genes. The resulting gene constructs were transfected into myeloma cells (Sp2/0) by electroporation Chinese hamster ovary lipofection. antibodies obtained exhibited same as that immunoglobulin produced hybridoma cell line. Antibody concentrations culture medium supernatants clonally but similar (15–480 ng/ml) both Sp2/0 transfectants; average production transfectants was only 3–5-fold less than transfectants. Ascites sufficiently high (900 µg/ml) initial studies with humanized T84.66.
