Semi-nondestructive genetic sampling from live eusocial wasps, Polistes dominulu and Polistes fuscatu

摘要: We present a method of extracting DNA from medium-sized paper wasps without sacrificing the animal. From distal half single leg, enough is extracted for 125 PCR reactions. This - collected Polistes dominulus and P. fuscatus– amplifies as well larger amounts tissue taken sacrificed animals, resulting microsatellite regions amplify faithfully. Although nest tenure workers with leg-cuts was shorter than control test animals appeared to behave normally. extraction suited studies that combine behavioral observations genetic analysis on likely disappear during observation period. Collecting data destroying colonies will enable researchers examine behavior in eusocial insects damaging future population size or altering structure.
