Identification of a pou2 ortholog in Chinese sturgeon, Acipenser sinensis and its expression patterns in tissues, immature individuals and during embryogenesis

摘要: The class V POU family genes, including pou5f1 and pou2, encode transcription factors critical for the maintenance of pluripotency in embryonic stem cells (ESC) germ line vertebrates. In present study, full-length cDNA a pou2 ortholog A. sinensis, Aspou2, was cloned sequenced. This sequence is 2,853 base pairs length encodes peptide 431 amino acid residues. A comparison deduced Aspou2 with that other vertebrate species showed they were highly conserved domain, which shared 88 90% identity zebrafish medaka, respectively, 69, 67 67% identical to frog, mouse human, respectively. RT-PCR analysis revealed detected all tissues examined except liver, high mRNA levels found muscle, pituitary brain. During embryogenesis early larval development, expression level mRNAs decreased gradually apart from 1-day larvae not observed. Furthermore, seemed raise development gonads immature Chinese sturgeons. These results suggested possible involvement nonpluripotent cells, pluripotent embryogenesis, gonad development.
