摘要: Fission yeast temperature-sensitive mutants cut3-477 and cut14-208 fail to condense chromosomes but small portions of the can separate along spindle during mitosis, producing phi-shaped chromosomes. Septation cell division occur in absence normal nuclear division, causing cut phenotype. Fluorescence situ hybridization demonstrated that contraction chromosome arm mitosis was defective. Mutant are apparently not rigid enough be transported poleward by spindle. Loss cut3 protein gene disruption fails maintain chromatin architecture even interphase. Both cut14 proteins contain a putative nucleoside triphosphate (NTP)-binding domain belong same ubiquitous family which includes budding Smc1 protein. The mutant suppressed an increase cut14+ dosage. protein, having highest similarity mouse is localized nucleus throughout cycle. Plasmids carrying DNA topoisomerase I partly temperature sensitive phenotype cut3-477, suggesting might involved topology.
