摘要: Abstract A rapid liquid chromatography/positive ion electrospray mass spectrometric assay (LC/ESI-MS) was developed for the quantitation of methotrexate, folinic acid, folic acid and ondansetron in human serum. The based on 100 μL serum samples, following acetonitrile precipitation proteins filtration that enabled direct injection into LC/MS system. All analytes internal standard, alfuzosin, were separated by using a Zorbax Eclipse XDB–C8 analytical column (2.1 mm × 150.0 mm i.d., particle size 3.5 μm) with isocratic elution. mobile phase composed mixture water/acetonitrile containing 0.1%, v/v formic (75:25, v/v), pumped at flow rate 0.15 mL min−1. Quantitation performed selected monitoring (SIM) positive ionization mode interface. found to be linear concentration range 0.01–25.00 μg mL−1 methotrexate 0.01–5.00 μg mL−1 ondansetron. Intermediate precision less than 4.2% over tested ranges. run time 7.0 min each sample made it possible analyze large number samples per day. method can used quantify covering variety clinical studies applied analysis obtained from children acute lymphoblastic leukemia.
