Differential oncogene amplification in tumor cells from a patient treated with cisplatin and 5-fluorouracil☆

摘要: Abstract Peritoneal cells were derived from a patient (PK) with adenocarcinoma of the colon during course cisplatin/ 5 -fluorouracil ( -FUra) treatment. Resistance to cisplatin and -FUra, characterized by lack response chemotherapy continued growth tumor, was concomitantly associated 2–4 -fold increase in DNA copy number for dTMP synthase dihydrofolate reductase. There corresponding amplification c- myc 2× ), H-ras 4× fos 15× ) oncogenes. Cytogenetic studies revealed an iso 13 q) chromosome, but failed show any double minutes or homogeneously staining regions. In addition, drug-resistant tumor PK another (HG) displayed enhanced expression synthase, polymerase β when compared normal tissue HCT 8 human carcinoma cell line. These results suggest that elevated oncogene gene may be involved development resistance.
