摘要: A cytomegalovirus (CMV)-specific sandwich hybridization test was constructed by using two adjacent BamHI DNA fragments of CMV as reagents. The were cloned into different vectors. One the recombinants attached to filter, and other labeled probe. When present in sample, mediated labeling filter hybridizing both filter-bound applied for detection from urine. released virus 2% Sarkosyl, concentrated 2-butanol extraction isopropanol precipitation, denatured, finally subjected test. As a result, 70 90% original viral could be recovered demonstrated quantitative reaction. Urine stored at room temperature Sarkosyl least 2 days without affecting detectability CMV. clinical applicability evaluated studying urine samples four infants excreting Sandwich presence all specimens. These contained originally 10(5) 10(8) molecules per ml.
