Effect of equilibration period on the viability of frozen-thawed mouse morulae after rapid freezing.

摘要: Mouse morulae were frozen with 1.5-4.0 M glycerol + 0.25 lactose solution by direct plunging into liquid nitrogen vapor 0.5-30 min after equilibration at room temperature. After thawing, embryos cultured in vitro, and the highest survival rates obtained exposure for 3 3.0 4.0 5 1.5 2.0 levels. Significant reductions (P less than 0.05) observed when periods extended 3-5 5-10 These results clearly demonstrate that time of glycerol-lactose mixture is one most important factors present rapid freezing conditions. To clarify lower embryo viability prolonged equilibration, we performed further experiments on effects to developmental potential without volume changes during or lactose. It was suggested detrimental are due not only toxicity osmotic injury higher concentrations cryoprotectant but also influx water embryonic cells caused hypotonic salt concentration extracellular (freezing) solution.
