High-Resolution Mapping of Gene Expression Using Association in an Outbred Mouse Stock

摘要: Quantitative trait locus (QTL) analysis is a powerful tool for mapping genes complex traits in mice, but its utility limited by poor resolution. A promising approach association outbred stocks or different inbred strains. As proof of concept the approach, we applied whole-genome to hepatic gene expression an mouse population, MF1 stock, and replicated QTL (eQTL) identified previous studies F2 intercross mice. We found that resolution these eQTL was significantly greater population. Through example, also showed how this precise can be used resolve previously loci (in studies), which affect many transcript levels (known as “hotspots”), into distinct regions. Our results highlight importance correcting population structure stock.
