摘要: Objective: Dopaminergic abnormalities in frontal-subcortical circuits have been hypothesized as the underlying pathophysiologic mechanism Tourette’s syndrome. The objective of this study was to test hypothesis that presynaptic dopamine release from striatum is abnormal adults with Method: Seven syndrome and five age-matched comparison subjects each received two positron emission tomography (PET) scans high specific activity [ 11 C]raclopride. first scan followed an intravenous injection saline; second amphetamine. relative estimated percentage difference binding potential between postsaline postamphetamine scans. Results: Binding determined after initial C]raclopride did not significantly differ subjects. After amphetamine challenge, mean value intrasynaptic putamen (as by true equilibrium bolus estimation) increased 21% change subjects; values 16.9% decreased 1.8%, respectively, when measured constrained method. Dopamine caudate region different Conclusions: Greater seen than a pharmacologic challenge These results suggest pathobiology phasic dysfunction transmission.
