摘要: Parathyroid hormone (PTH), PTH-related peptide (PTHrP), and calcium-sensing receptor (CaSR) play important roles in maintaining calcium homeostasis. Here, we study the effect of fluoride on expression PTH, PTHrP, CaSR both vitro vivo. MC3T3-E1 cells Sprague-Dawley rats were treated with different concentrations fluoride. Then, free ion concentration cell culture supernatant serum measured by biochemical analyzer. The was analyzed qRT-PCR Western blot. We found that low dose increased ionized (i[Ca2+]) high decreased i[Ca2+] supernatant. inhibited PTH PTHrP cells. improved Interestingly, NaF rats. Fluoride at messenger RNA (mRNA) protein levels fed regular diet low-calcium diet. also NF-kappaB ligand (RANKL), osteoprotegerin (OPG) ratio RANKL/OPG food presence or absence significantly increased. These results indicated might be able to affect homeostasis regulating CaSR.
