摘要: Inactivation of the p53 tumor suppressor is a frequent event in tumorigenesis. In most cases, gene mutated, giving rise to stable mutant protein whose accumulation regarded as hallmark cancer cells. Mutant proteins not only lose their suppressive activities but often gain additional oncogenic functions that endow cells with growth and survival advantages. Interestingly, mutations were shown occur at different phases multistep process malignant transformation, thus contributing differentially initiation, promotion, aggressiveness, metastasis. Here, authors review studies on involvement inactivation various stages tumorigenesis highlight specific contribution each phase progression.
