Cell growth and detachment from protein-coated PNIPAAm-based copolymers

摘要: The cultivation of cells requires the use unfavorable proteolytic enzymes, which cause cell-surface modification and also need considerable optimization. Recently, with development smart polymers, research has looked to using thermoresponsive polymers as cell culture substrates. These novel surfaces allow without enzymes by utilizing phase transition property poly(N-isopropylacrylamide) (PNIPAAm). Copolymers PNIPAAm N-tert-butyl-acrylamide (NtBAm) varying ratios were synthesized solvent cast. copolymer films are coated adhesion promoters such collagen, poly-L-lysine, laminin increase their growth properties. Cell activity measured alamarBlue™ PicoGreen® assays is similar for standard tissue plastic controls. Deposition onto was characterized scanning electron microscopy (SEM) atomic force (AFM). detachment from dependent on composition not affected surface coatings extracellular matrix (ECM) proteins. results demonstrate a versatile method while eliminating digestive trypsin. © 2007 Wiley Periodicals, Inc.
