The rapid diagnosis infectious diseases, particularly those that represent a public health problem, like tuberculosis, is challenging problem. By using nucleic acid amplification techniques PCR, one may be able to diagnose, the disease on day arrival specimen in laboratory. For tuberculosis by direct methods specimens from site infection are required. In certain cases it difficult get and such situations; some researchers have tried detect DNA Mycobacterium complex blood these patients. purposive this study determine diagnostic efficacy peripheral blood-based polymerase chain reaction for pulmonary tuberculosis. Methods. This was simple descriptive study, carried out Department Microbiology, Jan 2004 Dec 2004. Sputum samples were collected 96 suspected patients processed ZN staining AFB culture (gold standard) PCR. Results. Out cases, 60 (62.5%) positive. PCR positive 14 (14.5%). smear 34 (35.4%). overall sensitivity specificity assay 20% 94.4% respectively negative predictive values 85.71% 41.46% respectively. efficiency test 47.91%. Conclusion. Due low sensitivity; does not rule disease. However, helpful where available.
