Serine- and Threonine/Valine-Dependent Activation of PDK and Tor Orthologs Converge on Sch9 to Promote Aging

摘要: Dietary restriction extends longevity in organisms ranging from bacteria to mice and protects primates a variety of diseases, but the contribution each dietary component aging is poorly understood. Here we demonstrate that glucose specific amino acids promote stress sensitization through differential activation Ras/cAMP/PKA, PKH1/2 Tor/S6K pathways. Whereas sensitized cells Ras-dependent mechanism, threonine valine promoted cellular primarily by activating pathway serine via PDK1 orthologs Pkh1/2. Serine, activated signaling network which Sch9 integrates TORC1 Pkh phosphorylation threonines 570 737 intracellular relocalization transcriptional inhibition resistance protein kinase Rim15. Because conserved pro-aging role nutrient growth pathways higher eukaryotes, these results raise possibility similar mechanisms contribute mammals.
