摘要: A system has been developed for growth and maintenance of mammalian cells in suspension culture at high density. In principle, the constant levels required nutrients coupled with removal toxic cell byproducts can support much higher densities than may be obtained conventional spinners. The consisted 4- or 40-liter reaction vessels equipped a vertically supported rotating cylindrical filter. Agitation was provided by magnetically driven, Fresh medium supplied rate 10 to 20 ml/h per 109 expended free withdrawn through Both pH dissolved O2 CO2 were monitored regulated. Walker 256 carcinosarcoma have grown these reactors 10-to 30-fold greater that Bellco addition densities, yield liter used 2- 3-fold systems. 4-and versions this reactor operated without use antibiotics. also modified chemostat operation. single run, example, density maintained between 6 10×106 cells/ml total 8.7×1011 from 360 liters medium.
