摘要: Astilbin is a potential immunosuppressive agent with minor cytotoxicity. Its oral bioavailability supposed to be rather low and therefore sensitive analytical method required for its pharmacokinetic study after administration. A simple, rapid liquid chromatography-tandem mass spectrometry(LC-MS/MS) was developed validated the determination of astilbin in rat plasma. Plasma samples were subjected liquid-liquid extraction ethyl acetate separated by reversed phase high performance chromatography(HPLC) methanol-0.01%(volume fraction) formic acid(50:50, volume ratio) as mobile phase. Quantitive achieved on negative LC-MS/MS multiple reaction moitoring transitions m/z 449.1→150.9(quantifier) 449.1→284.9(qualifier) 128.9→42.0 internal standard(IS). lower limit quantification(LLOQ) ng/mL within short cycle time 3.4 min. The successfully applied involving intravenous administrations 6 mg/kg six rats.
