Catalytic Activity of Immobilized Chymotrypsin on Hybrid Silica-Magnetic Biocompatible Particles and Its Application in Peptide Synthesis

摘要: In this work, novel silica hybrid magnetic particles with biocompatible surface were designed as a support for enzyme immobilization, and the immobilized chymotrypsin (CT) performance was clarified model biocatalyst. CT is used in food technology drink clarification protein hydrolysis. The directly onto polydopamine-grafted (MNP@SiO2@PDA-CT) via Schiff base reaction. Immobilized had broadened both pH temperature profiles compared native CT. MNP@SiO2@PDA-CT system also improved thermostability enzyme. operated continuous reactor (CER) hydrolysis of different proteins (i.e., cytochrome c (Cyt c), human serum albumin (HSA), immunoglobulin G (HIgG), lysozyme (Lys)). peptide synthesis rate shown to decrease function increasing flow rate. catalytic activity CER remained stable 6.0 h operation period; thus, presented method may increase applicability area. showed good all tested proteins. peptides hydrolyzed from analyzed matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF-MS). Results show that permits area proteomic research.
