Optimization and hyper production of laccase from novel agaricomycete Pseudolagarobasidium acaciicola AGST3 and its application in in vitro decolorization of dyes

摘要: A novel white rot fungus Pseudolagarobasidium acaciicola AGST3 was investigated for the production of laccase under solid state fermentation. The effects fourteen medium components were screened by initial screening method Plackett-Burman. Each on basis a ‘p’ (probability value) that above 95 % confidence level. Tween 80, CuSO4·5H2O, glucose and FeC6H5O7·NH4OH identified as significant production. Central composite design using response surface methodology with four variables used in this study to optimize correlations between these optimum concentrations glucose, CuSO4·5H2O (g/l): 0.6625, 0.437, 0.037 respectively, maximum Compared unoptimized medium, (5.35 × 105 U/g substrate) increased 1.6-fold optimized condition. Furthermore, crude obtained decolorization structurally different dyes, 11–96 % after 24 h incubation.
