摘要: This study describes the development of a rapid and sensitive high-performance liquid chromatography–electrospray ionization tandem mass spectrometry (LC-MS/MS) assay for quantification [6]-gingerol in mouse plasma application to pharmacokinetic after dose ranging mice. The involved protein precipitation step with acetonitrile an isocratic elution using mobile phase consisting water containing 0.1% formic acid (80:20 v/v). multiple reaction monitoring was based on transition m/z = 277.2 177.1 294.2 137.1 nonivamide (internal standard). validated demonstrate specificity, linearity, recovery, accuracy, precision stability. calibration curves were linear over wide concentration range 10–10,000 ng/mL (r ≥ 0.9988). lower limit 10 ng/mL small volume (20 μL). method successfully applied mice intravenous injection at 1.5, 3 6 mg/kg doses. pharmacokinetics studied as demonstrated by increase area under concentration-time curve (AUCinf) no significant change systemic clearance (Cls), distribution (Vss) elimination half-life (t1/2) function dose.
