摘要: Dengue virus is the most prevalent mosquito-borne worldwide. In this study, we used pyrosequencing to analyze whole viral genome of two mouse-adapted strains, D2S10 and D2S20, that induce a dengue hemorrhagic fever/dengue shock syndrome (DHF/DSS)-like lethal disease in mice lacking type I and/or II interferon receptors. Previous experiments with indicated N124D K128E mutations envelope protein were responsible for severe induced compared its parental strain PL046. Here demonstrate D2S20 more virulent than captured presence five key amino acid – T70I, N83D, K122I (E), A62T nonstructural 2A (NS2A) G605V 5 (NS5) may account this. These findings set foundation further dissection determinants manifestations mouse models.
