摘要: Mitochondrial ribosomal RNAs (rRNAs) often display reduced size and deviant secondary structure, sometimes are fragmented, as their corresponding genes. Here we report a mitochondrial large subunit rRNA (mt-LSU rRNA) with unprecedented features. In the protist Diplonema ,t hernl gene is split into two pieces (modules 1 2, 534- 352-nt long) that encoded by distinct chromosomes, yet continuous. To reconstruct post-transcriptional maturation pathway of this rRNA, have catalogued transcript intermediates deep RNA sequencing RT-PCR. Gene modules transcribed separately. Subsequently, transcripts end-processed, module-1 polyuridylated module-2 polyadenylated. The joined via trans-splicing retains at junction 26 uridines, resulting in an extent insertion editing not observed before any system. A-tail trans-spliced molecules shorter than mono-module completely absent from mitoribosome-associated mt-LSU rRNA. We also characterize putative antisense transcripts. Antisense-mono-modules corroborate bi-directional transcription chromosomes. Antisense-mt-LSU if functional, has potential guiding concomitantly Together, these findings open window on investigation complex regulatory networks orchestrate multiple biochemically diverse events.
