Zirconium oxide aerogel for effective enrichment of phosphopeptides with high binding capacity

摘要: In this study, zirconium oxide (ZrO2) aerogel was synthesized via a green sol–gel approach, with oxychloride, instead of the commonly used alkoxide high toxicity, as precursor. With such material, phosphopeptides from digests 4 pmol β-casein coexistence 100 times (mol ratio) BSA could be selectively captured, and identified by MALDI-TOF MS. Due to large surface area (416.0 m2 g−1) mesoporous structure (the average pore size 10.2 nm) ZrO2 aerogel, 20-fold higher loading capacity for phosphopeptide, YKVPQLEIVPN[pS]AEER (MW 1952.12), obtained compared that commercial microspheres (341.5 vs. 17.87 mg g−1). The metal further applied in enrichment ng nonfat milk, 17 were positively identified, 1.5-fold improvement phosphopeptide detection previously reported results. These results demonstrate can powerful material phosphoproteome study.
