Augmenter of liver regeneration causes different kinetics of ERK1/2 and Akt/PKB phosphorylation than EGF and induces hepatocyte proliferation in an EGF receptor independent and liver specific manner.

摘要: Abstract Background/Aim Augmenter of liver regeneration (ALR) is a potent growth factor which supports in experimental animals. The aim this study was to compare proliferation as well the kinetics ERK1/2 and Akt/PKB phosphorylation by recombinant human ALR (rhALR) EGF hepatocytes extrahepatic cells. Methods Kinetics were determined primary (phh) after stimulation with rhALR EGF. Induction analyzed phh several cell lines hepatic origin MTT [3H]-thymidine assay. Results ERK showed clear differences, whereby caused transient permanent increase during observation period 60 min. For both, Akt phosphorylation, faster effect maximal levels observed already 2 min, whereas between 10 15 min. Using receptor inhibitor AG1478 we provide evidence an independent induction rhALR. Furthermore, induced only derived HepG2 Chang. In contrast, enhanced all types including colon, bronchial, pancreatic gastric (SW480, BC1, L36PL GC1). Conclusion induce different hepatocytes. mitogenic specific seems be at least partially from mediated signaling.
