l-selectin and α4β7 integrin, but not ICAM-1, regulate lymphocyte distribution in gut-associated lymphoid tissue of mice

摘要: Background Adhesion molecules on lymphocytes ( l -selectin and α4β7) endothelium (MAdCAM-1 ICAM-1) direct into the gut-associated lymphoid tissue (GALT) of mice. Parenteral nutrition MAdCAM-1 blockade reduce GALT cell mass. This study examined effects mass -selectin, α4β7, ICAM-1 with saturating doses monoclonal antibodies. Methods In experiment 1, α4β7 expression were measured by flow cytometry in chow-fed 2, 49 mice randomly received chow, parenteral nutrition, chow + intravenous (IV) anti-CD62L, chow + IV anti-LPAM-1, or isotype control antibody. After 4 days, lymphocyte yields respiratory intestinal IgA levels measured. 3, 27 anti-ICAM-1 antibody, antibody for 5 days. Lymphocyte counts determined as 2. Results Some 80% all circulating positive α4β7. Peyer's patches, lamina propria, intraepithelial space lower groups (3.1, 1.8, 0.9 × 10 6 2.1, 1.9, 0.7 , respectively) than chow group (5.9, 3.0, 1.7 ; P Conclusion Most have homing potential. Their distribution is hindered but not ICAM-1.
