Preliminary in vivo efficacy studies of a recombinant rhesus anti-α4β7 monoclonal antibody

摘要: Recent findings established that primary targets of HIV/SIV are lymphoid cells within the gastrointestinal (GI) tract. Focus has therefore shifted to T-cells expressing α4β7 integrin which facilitates trafficking GI tract via binding MAdCAM-1. Approaches better understand role α4β7+ in pathogenesis include their depletion or blockade synthesis, and/or homing capabilities vivo. Such studies can ideally be conducted rhesus macaques (RM), non-human primate model AIDS. Characterization expression on cell lineages RM blood and tissues reveal low densities by NK cells, B-cells, naive TEM (effector memory) T-cells. High were observed TCM (central Intravenous administration a single 50 mg/kg dose recombinant antibody resulted significant initial decline lymphocytes sustained coating receptor both periphery tissues.
