Capsaicin-induced apoptosis in SK-Hep-1 hepatocarcinoma cells involves Bcl-2 downregulation and caspase-3 activation.

摘要: Hepatocellular carcinoma is one of the most lethal malignancies and there no effective preventive measure in this highly malignant disease to date. In present study, we investigated chemopreventive potential capsaicin (8-methyl-N- vanillyl-6-nonenamide), principal pungent ingredient found hot red pepper, SK-Hep-1 hepatocellular cells. Treatment inhibited growth cells a concentration-dependent manner while 4-methoxy (Met-capsaicin) was less potent. This inhibitory effect on cell mainly due induction apoptosis as evidenced by DNA fragmentation nuclear condensation. Furthermore, prominently reduced ratio anti-apoptotic Bcl-2 pro-apoptotic Bax consequently increased caspase-3 activity. These results demonstrate that efficiently induced through caspase-3-dependent mechanism, which may contribute its function.
