摘要: The yellow lupin leghemoglobin I gene (lbI) was cloned in the pET-3a vector. It overexpressed Escherichia coli BL21(DE3)pLysS cells, under control of T7 RNA polymerase promoter. recombinant LbI protein, containing E. derived heme, purified to homogeneity using ion exchange and gel filtration chromatography. protein has spectral immunochemical properties identical isolated from root nodules. identity between expressed polypeptide native also supported by microsequencing analysis N-terminus protein.
