Synthetic peptide vaccine design: synthesis and properties of a high-density multiple antigenic peptide system

摘要: A convenient and versatile approach to the direct synthesis of a peptide-antigen matrix by solid-phase method is described. The called multiple antigen peptide system (MAP) it utilizes simple scaffolding low number sequential levels (n) trifunctional amino acid as core 2n antigens form macromolecule with high density final Mr 10,000. MAP model chosen for study was an octa-branching consisting made up three lysine eight terminals anchoring antigens. MAP, containing both peptides 9-16 acids, prepared in single method. Six different MAPs elicited specific antibodies rabbits mice, which five produced that reacted their corresponding native proteins. In rabbits, sera had considerably higher titer than from same anchored covalently keyhole limpet hemocyanin carrier. Thus, provided general, but chemically unambiguous, preparation carrier-bound predetermined reproducible structure might be suitable generating vaccines.
