摘要: Mitogen-inducible gene-6 (Mig-6) is a cytosolic multiadaptor protein that best known for its role as negative feedback regulator of epidermal growth factor receptor (EGFR) mediated signalling. Alternative roles Mig-6 are becoming increasingly recognised. Consistently with this, was demonstrated to be involved in broad spectrum cellular events including tumour suppression which may include the induction senescence. Here, we investigated mechanisms induced premature cell Endogenous poorly expressed young fibroblasts, whilst expression rises cells presenting typical features Overexpression sufficient trigger senescence early passage diploid lung fibroblasts (WI-38). Interestingly, overexpression reduced retinoblastoma (pRb) phosphorylation at inactivating Ser249/Thr252 sites. We also found these sites pRb increased presence B-Raf V600E oncogenic mutation. further show reduces inactivation and preserves function.
