Studying enzymatic bioreactions in a millisecond microfluidic flow mixer

摘要: In this study, the pre-steady state development of enzymatic bioreactions using a microfluidic mixer is presented. To follow such reactions fast mixing reagents (enzyme and substrate) crucial. By highly efficient passive micromixer based on multilaminar flow, times in low millisecond range are reached. Four lamination layers shallow channel reduce diffusion lengths to few micrometers only, enabling very mixing. This was proven by confocal fluorescence measurements channel’s cross sectional area. Adjusting overall flow rate 200 μm wide 900 μm long observation makes it possible investigate enzyme over several seconds. Further, device enables changing enzyme/substrate ratio from 1:1 up 3:1, while still providing high efficiency, as shown for hydrolysis β-galactosidase. way, early kinetics reaction at multiple concentrations can be collected short time (minutes). The easy handling powerful convenient instrument temporal analysis bioreactions.
