摘要: The mechanisms of rickettsial attachment have been studied by measuring quantitative changes in binding to host cells flow cytometry after different treatments the rickettsiae and cells. Time-dependent Rickettsia conorii was demonstrated increasing intensity cell surface fluorescence rickettsia-host combinations when examined with a rickettsia-specific monoclonal antibody. More than 70% had above threshold value 10 min incubation, owing bound surface, greatest indicative occurred at 20 min. kinetics dose dependent. greatly decreased or were treated 1% paraformaldehyde for 30 0.25% trypsin 5 15 min, respectively. Rickettsiae that heated 56 degrees C lost more 80% their ability attach R. rickettsii, an organism closely related conorii, competitively inhibited (51% inhibition mixed equal numbers). These results indicate structures are heat sensitive likely be proteins.
