Cyclin D2 is a moderately oscillating nucleoprotein required for G1 phase progression in specific cell types

摘要: To explore regulation and function of cyclin D2, a candidate cell cycle-regulatory proto-oncogene, we examined subcellular localisation, type- cycle-dependent expression, requirement D2 protein for G1 progression, in panel 40 human normal cancer types. Except lymphoid cells sarcoma lines, expression was considerably more restricted than that D1, whereas both D-type proteins were low or undetectable lacking functional retinoblastoma gene product. In cells, the resistant to extraction localised predominantly nuclei, it became soluble distributed nuclei cytoplasm from G1/S transition onwards. Centrifugal elutriation multiparameter flow cytometry analyses several types showed moderate cycle oscillation with maximum levels reached late G1. Microinjection and/or electroporation antibodies during arrested D2-expressing lymphocytes, breast myoepithelium, U-2-OS phase, D2-negative unaffected by such treatment.
