摘要: The temperature at which collagen denatures from a triple helix to random coil structure is useful measure of the degree crosslinking. A new multi-sample denaturation tester (DTT) has been constructed for rapid determination natural tissues and collagenous biomaterials. To validate system, temperatures measured DTT are compared with results differential scanning calorimetry (DSC). Data presented bovine pericardium in three states ranging 68 85 degrees C: fresh, or crosslinked glutaraldehyde epoxide reagent Denacol EX-512 poly (glycidyl ether). Denaturation by were not significantly different those (DSC); however, DSC onset systematically occurred slightly lower than that DTT. This result, seen only fresh tissue agreement earlier experiments using hydrothermal isometric tension (HIT) testing. By contrast, identical exogenously materials. Since transition was independent initial load, this variable may be chosen yield sharper force-temperature transitions given sample geometry. instrument allows accurate assessment multiple samples fraction time required other methods.
