An Empirical Approach to Sufficient Similarity: Combining Exposure Data and Mixtures Toxicology Data

摘要: When assessing risks posed by environmental chemical mixtures, whole mixture approaches are preferred to component approaches. toxicological data on mixtures as they occur in the environment not available, Environmental Protection Agency guidance states that toxicity from a considered "sufficiently similar" can serve surrogate. We propose novel method examine whether sufficiently similar, when exposure and study at least one representative available. define sufficient similarity using equivalence testing methodology comparing distance between benchmark dose estimates for both data-rich data-poor cases. construct "similar risk indicator"(SMRI) (analogous hazard index) similar linking with toxicology data. The methods illustrated pyrethroid occurrence collected child care centers (CCC) dose-response examining acute neurobehavioral effects of rats. Our shows 90% CCCs were mixture. Using hypothetical child, 95th percentile (weighted) SMRI these was 0.20 (i.e., where 1, more concern).
