DNA supercoiling factor contributes to dosage compensation in Drosophila

摘要: DNA supercoiling factor (SCF) is a protein capable of generating negative supercoils in conjunction with topoisomerase II. To clarify the biological functions SCF, we introduced heritable SCF RNAi into Drosophila. Upon knockdown observed male lethality and male-specific reduction expression levels X-linked genes. functionally interacts components MSL complex, which are required for dosage compensation via hypertranscription X chromosome. Moreover, colocalizes complex along overexpression chromosome had bloated appearance. This phenotype was dependent on histone acetyltransferase MOF suppressed by simultaneous ISWI. These findings demonstrate that plays role transcriptional activation alteration chromatin structure provide evidence contributes to compensation.
