摘要: Aim: The relative bioavailabilities and effects on lung injury alleviation of 4 insulin-artificial pulmonary surfactant (INS-APS) preparations were studied in normal rats. relationship between the minimal surface tension (γ min ) INS-APS absorption insulin was also investigated. Methods: Four formulations APS [1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC)/lecithin/palmitic acid (PA), DPPC/1-hexadecanol (Hex)/tyloxapol (Tyl), DPPC/L-α-phosphatidyl-DL-glycerol sodium salt (PG), DPPC/Tyl] prepared by thin-film sonication method direct sonication. γ dispersions examined with without INS pulsating a bubble tensiometer. In vivo experiments performed which serum glucose change level measured an enzymatic reagent kit radioimmunology assay after IT to reduction following 7 d consecutive administration evaluated edema index (the weight ratio wet dry lung) histopathology examination. Results: all below 10 mN/m. There no significant difference (P> 0.05) corresponding INS-APS. showed decrease increase (P INS/DPPC/Tyl, INS/DPPC/PG, control group. indices histopathological observation indicated that could alleviate injury. Conclusion: most potent hypoglycemic effect this study obtained INS/DPPC/Tyl. According results, there linear correlation bioavailability INS-APS, suggesting possible carriers insulin. APS, DPPC/Tyl, DPPC/PG might be efficient delivery achieving lower , enhancing absorption, decreasing injury.
