Serum-free media suitable for upstream and downstream processing

摘要: During the past decade use of serum-free media has become a standard method for cultivation most mammalian cell lines which are importance production biotechnologically relevant products such as biopharmaceuticals. More recently, there have been also successful efforts in development protein-free various lines. Ideally these should facilitate and simplify downstream processing, reduce costs minimize risk virus bacterial contamination. In addition, productivity several could be increased. However, not all support growth cells culture flasks suitable when used large-scale fermenters, expose to mechanical stress homogeneous conditions without microenvironments. Downstream processing is indeed simplified protein-poor or media, but lack protective carrier proteins leads increased product degradation by proteases released from cells, occurs even viable cultures. Thus, necessary compromises made between requirements upstream processes order an optimal quality.
