Using SERS-based microfluidic paper-based device (μPAD) for calibration-free quantitative measurement of AMI cardiac biomarkers

摘要: Abstract Recently, surface enhanced Raman scattering (SERS) has attracted much attention in medical diagnosis applications owing to better detection sensitivity and lower limit of (LOD) than colorimetric detection. In this paper, a novel calibration-free SERS−based μPAD with multi-reaction zones for simultaneous quantitative multiple cardiac biomarkers — GPBB, CK−MB cTnT early prognosis acute myocardial infarction (AMI) are presented. Three distinct probes were synthesised, subsequently conjugated respective detecting antibodies used as SERS nanotags biomarker Using conventional calibration curve, measurement on SERS-based was performed based the characteristic spectral features each reporter different nanotags. However, free point-of-care testing device is required fast screening rule-in rule-out AMI patients. Partial least squares predictive models developed incorporated into immunosensing system, accurately quantify three unknown levels serum previously obtained data. This method allows absolute when curve fails provide accurate estimation biomarkers, especially at low high concentration ranges. Under an optimised condition, LOD our identified 8, 10, 1 pg mL−1, cTnT, respectively, which well below clinical cutoff values. Therefore, proof-of-concept technique shows significant potential highly sensitive multiplex human expedite decisions patient care.
