摘要: Bone cells respond to mechanical stimuli, but the transduction mechanisms responsible are not fully understood. Integrins, a family of heterodimeric transmembrane glycoproteins, which link components extracellular matrix with actin cytoskeleton, have been implicated as mechanoreceptors. We assessed roles integrins in cyclical stimuli human bone (HBCs), results changes membrane potential. HBC showed depolarization following 0.104 Hz stimulation and hyperpolarization at 0.33 Hz. The response involved tetrodotoxin-sensitive sodium channels could be inhibited by antibodies against alpha V, beta 1, 5 integrins. In contrast, was gadolinium integrin-associated protein (CD47), 1 integrin. Both responses abrogated ARg-Gly-Asp (RGD)-containing peptides, inhibition tyrosine kinase activity, disruption cytoskeleton. These demonstrate differential electrophysiological different frequencies strain. Furthermore, they suggest that act mechanoreceptors distinct signaling pathways being activated stimuli.
