摘要: mRNA coding for rat liver mitochondrial aldehyde dehydrogenase was translated in rabbit reticulocytes 10 times more efficiently than the corresponding bovine mRNA. Previous data showed that mRNAs primarily differed noncoding 5'-end (K.-L. Guan and H. Weiner, manuscript submitted publication to Arch. Biochem. Biophys.). Fusion were constructed so 5'-untranslated region from one attached sequence of other. Furthermore, mutated at position -3, a implicated translational regulation, both contained same residues. Results indicated dramatic difference rates between just partly due around initiation codon. Secondary structure prediction stable secondary could exist mRNA, whereas no such should Existence predicted substantiated by observation termination DNA sequencing cDNA occurred. It is concluded performs major role inhibiting translation. This inhibitory effect related magnitude free energy required melt order reach AUG
