摘要: Summary This study aimed to compare the in vitro effects of coculture with Sertoli and SIM mouse embryo-derived thioguanine- ouabain-resistant (STO) feeder layer cells on efficiency adult spermatogonial stem (SSCs) colony formation. SSCs were isolated from testes, their identity was confirmed using immunocytochemistry against Oct4, CDH1, PLZF C-kit for vimentin cells. cultured a simple culture system (control group) top STO layers 2 weeks. The number diameter colonies evaluated during third, 7th, 10th 14th day culture, expression Oct-4, α6 β1 integrins assessed quantitative RT-PCR. Significant differences observed between three groups, separately each time (P < 0.05), higher mean (P < 0.05). results RT-PCR showed gene integrin group, but no significant Oct-4 among groups. Based optimal effect Seroli formation SSCs, it is suggested use these better colonisation SSCs.
