Effects of clomiphene on luteal function in the nonpregnant cynomolgus macaque.

摘要: Although estradiol-17 beta (E2) induces premature regression of the corpus luteum (CL), its role in spontaneous luteolysis which occurs at end nonfertile cycle has not been demonstrated. We compared effects an estrogen antagonist on E2-induced and by administering clomiphene (10 mg/day) to cynomolgus macaques during luteal phase presence absence exogenous E2 (supplied subcutaneous Silastic implants). Other animals received either vehicle or implants. Luteal function was assessed progesterone concentrations length. Clomiphene maintained normal luteolytic levels five six monkeys. However, alone did prolong beyond that observed monkeys receiving vehicle. To assess direct effect CL, we incubated monkey cells with human chorionic gonadotropin clomiphene, E2, plus E2. (1500 ng/ml) (1000 significantly (P less than 0.05) inhibited progestin production. together depressed production even greater extent. The data suggest mechanisms involved differ.
